Tosoh Bioscience a leading manufacturer of chromatographic resins introduces the new Toyopearl AF-rProtein A-650F affinity resin. It is well-suited for high capacity capturing of immunoglobulins out of high titre feedstocks. Toyopearl AF-rProtein-650F binds human and mouse immunoglobulin G (IgG), as well as IgM and Fab fragments. The resin combines a high hIgG dynamic binding capacity at short residence time and fast mass transfer kinetics with an improved alkaline stability. Thus, Toyopearl AF-rProtein A-650F offers the opportunity to improve process economics by increasing both, throughput and robustness.
Protein A based affinity chromatography is the industry standard for the capture of monoclonal antibodies (MAbs) in downstream processing. Rising fermentation titres have triggered the demand for resins that increase downstream capacity and efficiency. The new Toyopearl AF-rProtein A-650F capture resin was designed to meet these requirements. It provides a typical dynamic binding capacity for IgG of more than 30 mg/ml resin at two minutes residence time.
A newly developed recombinant ligand (non-animal-derived) was linked to the well proven methacrylic polymer backbone of Toyopearl media. The ligand has been engineered to maintain capacity even after repeated exposure to alkaline solution. Its multipoint attachment to the Toyopearl matrix further enhances chemical and thermal stability. In practice this pays off for a low level of protein A leaching and also for a high resistance to alkaline solutions employed in cleaning-in-place (CIP) procedures. Due to its rigid polymer matrix Toyopearl AF-rProtein A-650F also provides excellent pressure flow characteristics. It is therefore ideally suited for industrial scale purification of antibodies in large process columns.
